Le concept de biosécurité en pisciculture marine : prévention de l'encéphalite virale chez l'ombrine subtropicale (Sciaenops ocellatus) en Martinique ArchiMer
Adee, Fabrice.
In 1985, red drum (Sciaenops ocellatus) was introduced in Martinique in arder to replace sea bass fish farming. Nodavirus antibodies were detected in the broodstock at the IFREMER station in Robert. Nodavirus is responsible for viral nervous necrosis, a disease that is spreading quickly in worlwide marine aquaculture. A biosecurity process was initiated at the Robert hatchery to select seronegative breeders and produce nodavirus free larvae. This process, based on a serological follow-up of fish antibodies by ELISA, allowed a reduction of the number of seropositive fish. A complementary serological study revealed a low incidence of nodavirus contamination in nodavirus free larvae placed in sea cages on different production sites

Langoni,H.; Modolo,J. R.; Pezerico,S. B.; Silva,R. C.; Castro,A. P. B.; Silva,A. V. da; Padovani,C. R..
Toxoplasmosis is a zoonosis caused by Toxoplasma gondii, a coccidian protozoan of worldwide distribution. The seroprevalence in canine population can be an alternative for measuring T. gondii urban spreading. A total of 780 blood samples from dogs were collected, during the yearly anti-rabies campaign, carried out by the Department of Veterinary Hygiene and Public Health, School of Veterinary Medicine and Animal Husbandry (FMVZ), São Paulo State University, UNESP, together with the county health authorities, in August 1999. Using Indirect Fluorescent Antibody Test (IFAT) for detecting antibodies anti-T. gondii in the sera samples, we observed that 258 dogs (33.1%) were positive. The associations between the serological results and the epidemiological...

Langoni,H.; Da Silva,A.V.; Pezerico,S.B.; De Lima,V.Y..
Equine leptospirosis can present a non-symptomatic form, an acute clinical form, or even develop chronically, causing reproductive alterations, such as abortion and recurrent uveitis. Since the prevalence of leptospirosis in several countries and regions is widely reported, the objective of this study was to verify the prevailing equine leptospirosis in different regions of Brazil. Sera from 1402 blood samples from horses of different age, sex, breed, and purpose were examined. These samples came from southeastern and central west states of Brazil. The method utilized was the Microscopic Agglutination Test (MAT), with 12 different Leptospira serovars. From the sera tested, 754 (54%) were positive for one (385) or more (372) serovars. These results were...

American visceral Leishmaniasis: a case report J. Venom. Anim. Toxins incl. Trop. Dis.
Langoni,H.; Lucheis,S. B.; Da Silva,R. C.; Castro,A. P. B.; Paes,A. C..
Visceral leishmaniasis is a zoonotic disease caused by parasites of the Leishmania genus. Dog is the major source of infection to man, especially in urban areas. The authors report a case of visceral leishmaniasis in a pit bull female dog from Bocaina, São Paulo, Brazil. The animal presented clinical signs compatible with leishmaniasis, including skin lesions in the body and partial damage of the external ears. The indirect fluorescent antibody test (IFAT) demonstrated a titer of 1280, and promastigote forms of Leishmania sp were isolated by the culture of bone marrow puncture. Cytological analysis of the lymph node and smear of the bone marrow puncture revealed macrophages containing amastigote forms of Leishmania sp in their inner region. The test of...

Comparative serology of the marine fish pathogen Vibrio anguillarum OMA
Grisez, L.; Ollevier, F.P..
The different serotyping systems, based on thermostable O antigens, reported for Vibrio anguillarum and V. ordalii were compared by quantitative agglutination, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and subsequent silver staining or Western blotting (immunoblotting) of purified lipopolysaccharide (LPS), using polyclonal rabbit antisera. The results demonstrate that 16 different serotypes within V. anguillarum (designated 01 to 016) can be distinguished. Each of these serotypes is characterized by a distinct polysaccharide banding pattern, as revealed by silver-stained gels of purified LPS. The comparative analysis allowed a complete alignment of the different stereotypes for the...

Chicken serologic response to Salmonella enterica serotype Typhimurium assessed by Elisa Rev. Bras. Ciênc. Avic.
Oliveira,GH; Berchieri Jr,A; Montassier,HJ.
This study evaluated two enzyme-linked immunosorbent assays (ELISA) in the detection of chicken serologic response against Salmonella enterica sorotype Typhimurium. The assays have used as detecting antigen the soluble bacterial proteins of a non-flagellated strain of Salmonella Typhimurium (AgTM), and antibody conjugated to peroxidase or alkaline phosphatase. According to the results, optimal dilutions of antigen (concentration 5.49 mg/mL) and serum samples in both assays were 1:20,000 and 1:1,000, respectively. In such conditions, the ELISA/AgTM was able to detect serological response to Salmonella Typhimurium. Cross-reactions to Salmonella serotypes Gallinarum and Pullorum were seen, but not with other serotypes such as Enteritidis.

Assessment of serological response of chickens to Salmonella Gallinarum and Salmonella Pullorum by Elisa Rev. Bras. Ciênc. Avic.
Oliveira,GH de; Berchieri Júnior,Â; Montassier,HJ; Fernandes,AC.
This study was done to assess an enzyme-linked immunosorbent assay (ELISA) to detect chicken serological response against Salmonella enterica serotypes Gallinarum and Pullorum. The assays have used soluble bacterial proteins of Salmonella Gallinarum strain 9 (AgSG) as detecting antigen and peroxidase and alkaline phosphatase conjugates. According to the results, the antigen, sera and conjugate concentrations were optimized. In addition, the assay using alkaline phosphatase and peroxidase conjugates was helpful to distinguish positive serological reaction to serotypes Gallinarum and Pullorum from Enteritidis.

Freitas Neto,OC de; Carrasco,AOT; Raso,TF; Sousa,RLM; Berchieri Júnior,A; Pinto,AA.
Ratite farming of has expanded worldwide. Due to the intensive farming methods used by ratite producers, preventive medicine practices should be established. In this context, the surveillance and control of some avian pathogens are essential for the success of the ratite industry; however, little is known on the health status of ratites in Brazil. Therefore, the prevalence of antibodies against Newcastle Disease virus, Chlamydophila psittaci, Mycoplasma gallisepticum, Mycoplasma synoviae, and Salmonella Pullorum were evaluated in 100 serum samples collected from commercial ostriches and in 80 serum samples from commercial rheas reared in Brazil. All sampled animals were clinically healthy. The results showed that all ostriches and rheas were serologically...

Infectious bursal disease virus: case report and experimental studies in vaccinated and unvaccinated SPF chickens and commercial broiler chicks Rev. Bras. Ciênc. Avic.
Scanavini Neto,H; Ito,NMK; Miyaji,CI; Lima,E de A; Okabayashi,S; Corrêa,ARA; Eleutério,GC; Zuanaze,MA.
IBDV Gm 11 (Simbios eleven-molecular group) has been detected since 1997 in many farms of commercial broilers and layers causing high mortality (2 to 15%) and severe macro and microscopic damage in cloacal bursae, spleen, thymus, kidney and liver. Five serial passages of 2050/97-Gm 11 IBDV sample by CAM route in SPF chicken's embryonated eggs did not elicit increased embryo mortality. High mortality (100%) of 21 day-old SPF leghorn chickens and severe bursal and splenic lesions were seen from 24 up to 48 hours after eye-drop inoculation of 2050/97 strain (50 mL of 10-2 dilution of 10% bursae homogenate). Mortality was not detected when vaccinated SPF and broiler chickens were inoculated. One dead bird was found among ten challenged unvaccinated broilers....

Test profiles of broiler breeder flocks housed in farms with endemic Mycoplasma synoviae infection Rev. Bras. Ciênc. Avic.
Fiorentin,L; Mores,MAZ; Trevisol,IM; Antunes,SC; Costa,JLA; Soncini,RA; Vieira,ND.
There is a need for a better understanding of the epidemiology of Mycoplasma synoviae (MS) infection in broiler breeders in Brazil. Many features of the infection remain unrecognizable, because there are no clinical signs of the disease. A detailed testing was performed at each 6 to 8 weeks in three MS-free flocks introduced in farms with endemic MS infection for a follow-up epidemiological study. Every flock was monitored by polymerase chain reaction (PCR), by serum plate agglutination (SPA) and hemagglutination inhibition (HI) for serology studies, and isolation of mycoplasmas from tracheal swabs. PCR was found to be the most sensitive test, detecting early MS infection. Serology was positive in less than 50% of the sera and MS was isolated only between...

Elisa evaluation of the levels of antibodies against Infectious Bronchitis Virus in laying hens using egg yolk as substrate Rev. Bras. Ciênc. Avic.
Rauber,RH; Flôres,ML; Pereira,CE; Grigulo,M.
The present work was carried out to compare Infectious Bronchitis Virus (IBV) antibody titers in serum and egg yolk samples from laying hens. Sixty paired blood and egg samples were collected from laying hens of two farms. Serum samples were frozen, while egg yolk samples were diluted (1:500) before freezing. Serum and yolk samples were tested for the presence of IBV antibodies by indirect ELISA (commercial kit) and titers were compared by a correlation test (alpha=0.05). There was a high correlation (r=0.62) between the two kinds of samples, which means that titers of IBV antibodies in the egg yolk and in serum samples are quite the same. Considering that blood collection causes deep stress that leads to economic losses, and since eggs are collected daily...
